Process Workflow

For 5 hours each time, the extract was filtered and concentrated under reduced pressure to obtain 8.3 kilograms of extract; dissolve the extract with 360 liters of methylene chloride and pour it into the extraction tank. Another 17.28 kilograms of NaOH was weighed to prepare 720 liters of 0.6moL/L NaOH solution. Put it into the extraction tank in two batches, wash the dichloromethane solution with alkali twice, and concentrate the dichloromethane phase to obtain 2 kilograms of alkali-washed extract; take 10 kilograms of 200-300 mesh Qingdao marine column chromatography normal phase silica gel, Wet method is used to install a column with a diameter of 35cm. After alkali washing, the extract is loaded and eluted with methylene chloride first. The eluate containing triptolide was collected, concentrated to about 1/8 volume and then left to crystallize at 4°C to obtain 13.8g of triptolide crude product; the crude product was reconstituted at room temperature using isopropyl ether as the crystallization solvent. After crystallization, 12.3g of pure triptolide was obtained by filtration, and its purity was 99.4% as detected by HPLC.
